Everolimus plus bevacizumab is a combination therapy used primarily in the treatment of advanced renal cell carcinoma, including both clear cell and papillary variants. Everolimus is an oral small molecule inhibitor of the mammalian target of rapamycin (mTOR), which blocks a key pathway involved in cancer cell growth, proliferation, and angiogenesis. Bevacizumab is a monoclonal antibody that targets vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis—the formation of new blood vessels that supply tumors. The combination provides dual inhibition of tumor growth and blood vessel formation, leading to improved progression-free survival in clinical trials for metastatic renal cell carcinoma. This regimen has demonstrated robust activity as first-line therapy for papillary variant RCC and has also shown efficacy in clear cell RCC, both as initial treatment and after prior targeted therapies[1][4][5][6].
