Overview

Everolimus + cisplatin is a combination regimen used in oncology clinical trials for the treatment of various advanced cancers. Everolimus is an mTOR inhibitor that binds to FKBP-12 and inhibits the activation of mTOR (mechanistic target of rapamycin), leading to reduced cell proliferation, angiogenesis, and glucose uptake by blocking progression from G1 to S phase in the cell cycle and inducing apoptosis[1][3][5]. Cisplatin is a platinum-based chemotherapy agent that acts as an alkylating agent; it cross-links DNA at guanine bases causing DNA damage and triggering apoptosis through multiple signaling pathways including p53 activation[2][6]. The combination has been studied in diseases such as advanced neuroendocrine carcinoma (EP-NEC), urothelial bladder cancer, and small cell lung cancer[3][5][8].
