Overview

**Everolimus + cisplatin + etoposide** is a three-drug combination therapy investigated primarily for the first-line treatment of patients with extensive-stage small-cell lung cancer (SCLC). Everolimus is a small molecule inhibitor of the mammalian target of rapamycin (mTOR), which blocks mTORC1 kinase activity to inhibit cell growth, survival, and angiogenesis. Cisplatin is a platinum-based chemotherapeutic that induces DNA crosslinking and apoptosis, while etoposide inhibits topoisomerase II, preventing DNA replication and promoting cell death. This regimen is designed to combine targeted growth pathway inhibition via everolimus with cytotoxic effects from cisplatin and etoposide, aiming to improve tumor control over conventional platinum/etoposide doublet chemotherapy in SCLC and potentially other solid tumors[4][5].
