Overview

This drug is a combination regimen of three agents used in cancer treatment: - **Everolimus** is an mTOR inhibitor that blocks the mammalian target of rapamycin pathway, which is involved in cell growth and proliferation. - **Ganitumab** (AMG 479) is a monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), inhibiting its signaling which can promote tumor growth. - **Panitumumab** is a fully human monoclonal antibody against the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling pathways that drive cancer cell proliferation. The combination was studied primarily in advanced solid tumors including refractory non-small cell lung cancer and sarcoma. The triplet regimen showed unacceptable toxicity leading to discontinuation; however, the doublet of ganitumab plus everolimus had an acceptable safety profile with notable clinical activity. Panitumumab addition increased toxicity significantly[1][2][3].
