Overview

A combination drug of **everolimus** (a mammalian target of rapamycin [mTOR] inhibitor) and **lanreotide** (a synthetic somatostatin analogue) developed and studied specifically for the treatment of **unresectable or recurrent grade 1 or 2 gastroenteropancreatic neuroendocrine tumors (GEP-NETs) with poor prognostic factors**, such as high Ki-67 index or diffuse liver metastases[1][2][3][4][5][8]. Everolimus acts by inhibiting the mTOR pathway, suppressing tumor cell proliferation and growth. Lanreotide binds somatostatin receptors (mainly SSTR2 and SSTR5), inhibiting the secretion of various hormones and growth factors, and exerting direct antiproliferative effects on neuroendocrine tumor cells. In the phase III STARTER-NET trial, the combination therapy extended progression-free survival and improved objective response rates compared to everolimus alone, with adverse events generally manageable and a safety profile consistent with each drug's known profile[1][2][3][4][5][6][8]. The regimen may become a new standard of care for high-risk, aggressive, well-differentiated GEP-NETs.
