Overview

This is a combination of two investigational small molecule drugs—everolimus and linsitinib—studied primarily in oncology. Everolimus is an orally available inhibitor of the mammalian target of rapamycin (mTOR), a serine/threonine kinase involved in cell growth, proliferation, and survival. Linsitinib (OSI-906) is a potent dual inhibitor of insulin-like growth factor 1 receptor (IGF-1R) kinase and insulin receptor (InsR) kinase. The rationale for combining these agents is to simultaneously block mTOR signaling and IGF/Insulin pathways, which are often implicated in cancer cell survival and resistance mechanisms. This combination has been evaluated in early-phase clinical trials for refractory metastatic colorectal cancer but did not demonstrate significant clinical activity[1][3]. Both drugs have also been explored as part of multi-drug regimens targeting other cancers with aberrant PI3K/mTOR/IGF signaling[2][4].
