Overview

The combination of **everolimus** and **mycophenolic acid** is an immunosuppressive regimen primarily used to prevent organ rejection in kidney transplant recipients. **Everolimus** is a selective inhibitor of the serine/threonine-protein kinase mTOR, inhibiting cell growth, proliferation, and angiogenesis by arresting cells in the G1 phase and reducing downstream mTOR signaling[3]. **Mycophenolic acid** inhibits inosine monophosphate dehydrogenase (IMPDH), selectively blocking de novo guanine nucleotide synthesis essential for the proliferation and maturation of T and B lymphocytes[2][6]. Together, these agents exert a complementary and potent suppression of both cellular and humoral immune responses, reducing the risk of transplant rejection[7]. Typically this combination is used in patients at risk for or experiencing intolerance to calcineurin inhibitors or standard antimetabolite therapy[7].
