Overview

Everolimus + octreotide is a **combination therapy** consisting of everolimus, an oral small molecule inhibitor of the mammalian target of rapamycin (**mTOR**), and octreotide, a synthetic long-acting analogue of the hormone somatostatin, administered as octreotide LAR (long-acting repeatable) by intramuscular injection[1][2][3][4]. Everolimus inhibits mTOR signaling, thereby suppressing tumor cell growth, proliferation, and angiogenesis. Octreotide binds to somatostatin receptors (primarily subtype 2), inhibiting the secretion of various hormones and growth factors and exerting antiproliferative effects on neuroendocrine tumor cells. This combination is primarily indicated for the treatment of patients with advanced, well-differentiated neuroendocrine tumors (NETs) of gastroenteropancreatic (GEP) origin or lung. Clinical studies demonstrate improved progression-free survival and disease control versus either agent alone, with a manageable safety profile. The combination is supported and developed in part by Novartis[1][3].
