Everolimus + osimertinib is a combination therapy that pairs two distinct targeted cancer drugs. Everolimus is an mTOR inhibitor that specifically targets mTORC1, while osimertinib is a third-generation EGFR tyrosine kinase inhibitor (TKI) that targets specific EGFR mutations (T790M, L858R, and exon 19 deletions) common in non-small cell lung cancer (NSCLC). This combination has been investigated as a potential strategy to overcome resistance to EGFR-TKIs in NSCLC patients with acquired PI3K-AKT-mTOR pathway alterations. Clinical studies have shown that patients receiving this combination achieved stable disease with a progression-free survival of approximately 2.1-4.2 months, though treatment-related adverse events including oral ulcers and rash have been reported.
