everolimus + pemetrexed

Everolimus + pemetrexed is an investigational combination therapy consisting of everolimus, an mTOR (mammalian target of rapamycin) inhibitor, and pemetrexed, a folate antimetabolite chemotherapy agent. Everolimus inhibits the mTOR pathway, which regulates cell growth and proliferation. Pemetrexed inhibits several enzymes involved in purine and pyrimidine synthesis, primarily thymidylate synthase, thereby disrupting DNA replication in rapidly dividing tumor cells. This combination has been studied primarily for advanced non-small cell lung cancer (NSCLC), particularly after failure of prior chemotherapy regimens. Clinical studies have evaluated its safety profile and preliminary efficacy in this setting[1][2][5].
