Overview

This is a combination of two pharmaceutical drugs, everolimus and valganciclovir, used primarily in the context of organ transplantation. Everolimus is an mTOR (mechanistic target of rapamycin) inhibitor that acts as an immunosuppressant to prevent organ rejection and has demonstrated additional antiviral effects against cytomegalovirus (CMV), likely by interfering with viral protein synthesis through inhibition of the mTOR signaling pathway. Valganciclovir is an oral prodrug of ganciclovir, a guanine analog antiviral agent that requires phosphorylation by viral kinase UL97 for activation and inhibits CMV DNA polymerase (UL54), thereby blocking viral replication. The combination has been studied for prophylaxis against CMV infection in transplant recipients, with evidence suggesting superior efficacy compared to other strategies for preventing CMV infection after heart transplantation[1][4][6].
