Everolimus + vinorelbine is a combination of two small molecule anticancer drugs used primarily in the treatment of advanced or metastatic breast cancer, particularly HER2-positive cases that are resistant to trastuzumab and have been pre-treated with taxanes. Everolimus is an mTOR (mechanistic target of rapamycin) inhibitor that blocks a key signaling pathway involved in cell growth, proliferation, and survival. Vinorelbine is a semi-synthetic vinca alkaloid that inhibits microtubule assembly, thereby disrupting mitosis and inducing apoptosis in rapidly dividing cells. The combination targets multiple pathways to overcome resistance mechanisms and improve therapeutic outcomes. Clinical trials have also explored this combination for other cancers such as renal cell carcinoma[3][5][6].
