Evexomostat + 5-fluorouracil is an investigational combination therapy developed by SynDevRx for the treatment of metabolic hormone-sensitive melanoma. Evexomostat (SDX-7320) is a first-in-class, polymer-conjugated small molecule inhibitor of methionine aminopeptidase 2 (MetAP2). By irreversibly binding to MetAP2, evexomostat modulates metabolic hormones such as insulin, leptin, and adiponectin, while also inhibiting angiogenic factors like bFGF and VEGF-C. 5-fluorouracil is a classic antimetabolite chemotherapy that inhibits thymidylate synthase. This combination aims to address both the tumor biology and the systemic metabolic dysfunction often associated with cancer progression. It is currently in Phase 1 clinical development.
