Evexomostat + AKT inhibitor is a combination therapy under development by SynDevRx, primarily for the treatment of HER2-positive metastatic breast cancer. The combination consists of evexomostat (SDX-7320), a small molecule inhibitor of methionine aminopeptidase 2 (MetAP2), and an unspecified AKT inhibitor. Evexomostat is designed to modulate metabolic and angiogenic drivers of tumor growth, such as insulin, leptin, and adiponectin, which are often dysregulated in cancer patients and can contribute to treatment resistance. By inhibiting MetAP2, evexomostat aims to normalize these metabolic factors and inhibit angiogenesis. The addition of an AKT inhibitor targets the PI3K/AKT/mTOR signaling pathway, a critical survival pathway that is frequently hyperactivated in breast cancer. This dual approach is intended to provide synergistic anti-tumor activity while potentially mitigating metabolic side effects, such as hyperglycemia, that are commonly associated with AKT inhibition.
