Overview

Evorpacept + pembrolizumab is an investigational combination therapy under clinical development for the treatment of advanced solid tumors, most notably recurrent or metastatic head and neck squamous cell carcinoma (HNSCC). **Evorpacept** (code name ALX148) is a high-affinity CD47-blocking fusion protein with an inactive Fc region designed to block the "don't eat me" signal on cancer cells, thereby enhancing phagocytosis and activation of dendritic cells, and promoting both innate and adaptive immune activation. **Pembrolizumab** (brand name Keytruda; code name MK-3475) is an approved immune checkpoint inhibitor targeting PD-1, relieving PD-1/PD-L1-mediated inhibition of activated T cells to enable antitumor immune activity. The combination is designed to bridge innate and adaptive immune responses, with the goal of improving anti-tumor efficacy in solid tumors, especially HNSCC. This therapy is being developed primarily by ALX Oncology (evorpacept) and Merck (pembrolizumab)[2][3][4][1].
