Overview

A combination therapy of **evorpacept**, a recombinant fusion protein that acts as a CD47-blocking myeloid checkpoint inhibitor, and **trastuzumab**, a monoclonal antibody targeting the human epidermal growth factor receptor 2 (**HER2**). Evorpacept inhibits the "don't eat me" signal delivered by CD47 to SIRPα on macrophages, thereby enhancing anti-tumor phagocytosis and immune response. Trastuzumab binds HER2, inhibiting proliferation of HER2-overexpressing cancer cells. The combination is primarily developed for use in HER2-positive advanced solid tumors, in particular gastric and gastroesophageal junction (**GEJ**) cancers. Clinical studies demonstrated that evorpacept + trastuzumab shows antitumor activity, is well-tolerated, and is being pursued for further clinical development.
