Evorpacept + trastuzumab + eribulin is a combination therapy under investigation for cancer, particularly HER2-positive breast cancer and potentially other solid tumors. Evorpacept is a CD47-blocking fusion protein designed to enhance innate immune-mediated clearance of tumor cells by disrupting the “don’t eat me” signal that cancer cells use to evade immune destruction. Trastuzumab is a monoclonal antibody that targets the HER2 receptor, inhibiting proliferation of HER2-overexpressing tumor cells. Eribulin is a synthetic non-taxane microtubule inhibitor with both anti-mitotic and non-mitotic activities; it induces apoptosis and modulates the tumor microenvironment. The combination is designed to provide complementary anti-tumor activities: immune-mediated clearance (evorpacept), HER2 signaling inhibition (trastuzumab), and cytotoxic activity (eribulin). Clinical development includes evaluation in HER2-positive and other breast cancers, especially in patients who have received prior lines of therapy[1][2][5].
