Overview

**evorpacept + trastuzumab + vinorelbine** is an investigational combination therapy being evaluated for the treatment of metastatic HER2-positive breast cancer. **Evorpacept (ALX148)** is a high-affinity CD47-blocking fusion protein that enhances macrophage-mediated phagocytosis of tumor cells by inhibiting the "don't eat me" signal of CD47; it is designed to augment the anti-tumor effects of other agents. **Trastuzumab** is a monoclonal antibody targeting HER2 (human epidermal growth factor receptor 2) on cancer cells, blocking HER2-driven signaling and activating antibody-dependent cellular cytotoxicity. **Vinorelbine** is a chemotherapy drug of the vinca alkaloid class that interferes with microtubule formation during mitosis, inhibiting cancer cell division and growth. This three-drug regimen is being studied for its synergistic potential in advanced breast cancer, especially after prior trastuzumab-deruxtecan therapy, aiming to combine enhanced immune-mediated clearance with targeted inhibition and cytotoxicity[5][7].
