EVX-01 + pembrolizumab is a **combination investigational therapy** for unresectable or metastatic melanoma, currently in clinical development. **EVX-01** is a personalized peptide-based neoantigen vaccine designed using artificial intelligence algorithms (PIONEER platform) to target tumor-specific neoepitopes that are unique to each patient's cancer, stimulating the immune system by inducing potent, neoepitope-specific CD4+ and CD8+ T-cell responses. **Pembrolizumab** is a licensed, anti-PD-1 monoclonal antibody (checkpoint inhibitor) that restores T cell activity by blocking the programmed cell death protein 1 (PD-1) receptor. The rationale for this combination is to synergistically boost immune targeting of melanoma by both potentiating tumor-specific T-cell priming (via EVX-01) and preventing T-cell inhibition (via pembrolizumab). EVX-01 is administered intramuscularly, beginning after the initiation of pembrolizumab.
