Overview

EXL1 is a combination therapy consisting of the aromatase inhibitor **exemestane** and the multikinase inhibitor **lenvatinib**. Developed by the **Centre Hospitalier Universitaire de Lille** (CHU Lille), it is currently being evaluated in a Phase 2 clinical trial (NCT04653142) for patients with advanced non-small cell lung cancer (NSCLC) who have progressed on or are refractory to immune checkpoint inhibitor (ICI) therapy. The rationale for this combination lies in the potential synergistic effects of targeting the estrogen signaling pathway, which has been implicated in lung cancer progression and immune evasion, alongside multiple receptor tyrosine kinases (including VEGFR, FGFR, and PDGFR) involved in angiogenesis and tumor growth. This dual approach is intended to overcome resistance mechanisms in lung adenocarcinoma.
