Overview

Exemestane + seribantumab is an investigational combination therapy studied primarily in hormone receptor-positive, HER2-negative metastatic breast cancer. Exemestane is a steroidal aromatase inhibitor that irreversibly blocks the conversion of androgens to estrogens, thereby reducing estrogen levels and limiting the growth of estrogen-dependent tumors[4]. Seribantumab is a fully human IgG2 monoclonal antibody targeting the human epidermal growth factor receptor 3 (HER3/ErbB3). It antagonizes HER3 signaling by blocking heregulin (NRG1) binding, inhibiting downstream PI3K-AKT pathway activation and tumor cell proliferation[1][5][6]. The combination was evaluated in phase 2 clinical trials for postmenopausal women with ER/PR+, HER2– metastatic breast cancer. Exploratory analyses suggest patients with detectable HRG mRNA may benefit most from this regimen[2][3].
