Drug intelligence / Profile preview
exendin-4 + gastrin + xenin-8-Gln is an experimental, synthetic, triple-acting hybrid peptide designed for the treatment of type 2 diabetes and obesity. Developed by academic researchers at Ulster University, the chimeric peptide is constructed by linking the first 28 N-terminal amino acids of the GLP-1 receptor agonist exendin-4, gastrin-6, and an enzymatically stable xenin-8-Gln analogue using two 8-amino-3,6-dioxaoctanoic acid linkers. It acts as a tri-agonist targeting the glucagon-like peptide-1 (GLP-1) receptor, the cholecystokinin B (CCK2/gastrin) receptor, and the neurotensin receptor 1 (NTSR1, the receptor for xenin). Preclinical studies in diabetic and obese mouse models have demonstrated that the peptide significantly enhances insulin secretion, improves glucose tolerance and insulin sensitivity, reduces food intake, and promotes pancreatic beta-cell proliferation while reducing apoptosis.
