Overview

Exendin-9,39 is a peptide-based biologic that acts as a potent and competitive antagonist of the glucagon-like peptide-1 (GLP-1) receptor. It is a truncated version of exendin-4, lacking the N-terminal amino acids necessary for receptor activation, which allows it to bind to the GLP-1 receptor with high affinity without triggering a biological response. In clinical research, particularly studies led by Dr. Adrian Vella at the Mayo Clinic, it is used as a pharmacological tool to block endogenous GLP-1 signaling to evaluate its contribution to insulin secretion, glucose metabolism, and islet function. The compound has a short plasma half-life of approximately 30 minutes and is typically administered via intravenous infusion. While it has been utilized in Phase 3 research settings to understand metabolic physiology in patients with type 2 diabetes and obesity, it is primarily an investigational research tool rather than a marketed therapeutic agent.
