F16-IL2 + doxorubicin is a combination therapy under clinical investigation for the treatment of advanced solid tumors and metastatic breast cancer. F16-IL2 is a recombinant fusion protein, known as an immunocytokine, composed of the antibody fragment F16—which targets the A1 domain of tenascin-C in tumor stroma—and human interleukin 2 (IL2). This design enables selective localization to tumor tissues and enhances immune-mediated antitumor activity. Doxorubicin is an anthracycline chemotherapeutic agent that works primarily by intercalating into DNA and inhibiting topoisomerase II, leading to DNA damage and apoptosis in rapidly dividing cells. The combination aims to leverage both targeted immune activation via IL2 at the tumor site and direct cytotoxicity from doxorubicin. Clinical trials have shown this regimen can be safely administered with manageable toxicity profiles[5][4][3]. Philogen has been involved in developing F16-IL2[2].
