Overview

F16IL2 + nivolumab is an experimental **combination therapy** comprising the antibody-cytokine fusion protein F16IL2 and the anti-PD-1 monoclonal antibody nivolumab. **F16IL2** is a recombinant fusion protein consisting of a human monoclonal antibody fragment (F16) targeting the A1 domain of tenascin-C (an extracellular matrix glycoprotein upregulated in tumors) fused to human interleukin-2 (IL-2). It acts by anchoring IL-2 in the tumor microenvironment, stimulating immune cells such as NK cells and T cells at the disease site. **Nivolumab** is an immune checkpoint inhibitor against programmed cell death-1 (PD-1), enhancing T cell-mediated antitumor responses by preventing PD-1-mediated inhibition of immune activation. This combination aims to potentiate antitumor immunity by both stimulating cytotoxic lymphocytes (via IL-2) and releasing immune suppression (via PD-1 blockade). The combination is being evaluated for treatment of non-small cell lung cancer and has also been studied in other tumor types[1][3][5][6].
