Overview

F901318 (olorofim) is a novel antifungal agent belonging to the orotomide class, which inhibits fungal dihydroorotate dehydrogenase (DHODH), a key enzyme in the de novo pyrimidine biosynthesis pathway. Posaconazole is a triazole antifungal that inhibits lanosterol 14α-demethylase (CYP51A), disrupting ergosterol synthesis in fungal cell membranes. The combination of F901318 and posaconazole has been studied for potential additive or synergistic effects against invasive fungal infections, particularly those caused by Aspergillus species and other resistant fungi. Both drugs have demonstrated efficacy in preclinical models of invasive pulmonary aspergillosis, with F901318 showing activity against azole-resistant strains[1][4][6].
