Overview

Fab'CD3-MORF2 is a T-cell engaging component of the Multi-Antigen T-Cell Hybridizers (MATCH) system, a modular platform designed for the in situ assembly of bispecific T-cell engagers. It consists of an anti-CD3 antibody Fab' fragment covalently conjugated to a 25-base morpholino oligonucleotide (MORF2). The MATCH system functions through a pre-targeting strategy: a cancer-targeting Fab' fragment conjugated to a complementary oligonucleotide (MORF1) is first administered to bind to malignant cells. Subsequent administration of Fab'CD3-MORF2 allows the two components to hybridize on the cell surface, forming a functional bispecific complex. This complex recruits and activates T cells to induce targeted cytotoxicity against the cancer cells. This approach is being developed primarily for the treatment of multiple myeloma to overcome limitations of traditional bispecific antibodies, such as off-target toxicity and manufacturing complexity.
