Overview

Fab'CD33-MORF1 is a tumor-targeting antibody fragment conjugate developed by the University of Utah as part of the Multi-Antigen T Cell Hybridizers (MATCH) platform. It consists of a Fab' fragment of an anti-CD33 monoclonal antibody covalently conjugated to a 25-base morpholino oligonucleotide (MORF1). Fab'CD33-MORF1 is designed to target CD33-expressing cancer cells, such as those in acute myeloid leukemia (AML) and multiple myeloma (MM). When administered alongside a complementary T-cell-engaging module, such as Fab'CD3-MORF2, the programmable hybridization between the complementary morpholino oligonucleotides (MORF1 and MORF2) brings the tumor cells and T cells into close proximity. This in situ post-assembly of the bispecific complex facilitates immune synapse formation and potent, antigen-dependent T-cell-mediated cytotoxicity while allowing for tunable cytokine release and reduced toxicity compared to conventional bispecific T-cell engagers.
