Fab'CD28-MORF2 is a preclinical-stage, modular immune-engaging conjugate developed as part of the Multi-Antigen T Cell Hybridizers (MATCH) platform. It consists of an antigen-binding fragment (Fab') of an anti-CD28 monoclonal antibody covalently conjugated to a 25-base single-stranded morpholino oligonucleotide (MORF2). In the MATCH system, Fab'CD28-MORF2 acts as a modular costimulatory T-cell engager. When combined with a tumor-targeting Fab'-MORF1 conjugate (such as Fab'BCMA-MORF1 or Fab'PD-L1-MORF1) and a complementary T-cell-engaging Fab'CD3-MORF2 conjugate, the complementary MORF1 and MORF2 oligonucleotides hybridize via Watson-Crick base pairing. This self-assembly brings the tumor cells and T cells into close proximity, forming a functional immune synapse. The incorporation of Fab'CD28-MORF2 provides a costimulatory signal through CD28, which significantly enhances T-cell activation, proliferation, and tumor cell killing (especially in solid tumors with low antigen density or immunosuppressive microenvironments) while allowing precise control over cytokine release.
