This combination therapy consists of **farletuzumab**, a humanized monoclonal antibody targeting folate receptor alpha; **carboplatin**, a platinum-based chemotherapy; and **pegylated liposomal doxorubicin**, an anthracycline antineoplastic agent encapsulated for improved delivery and reduced toxicity. Farletuzumab exerts anti-tumor effects through immune-mediated mechanisms such as antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity by binding folate receptor alpha which is overexpressed in epithelial ovarian cancer. Carboplatin induces DNA crosslinks leading to apoptosis. Pegylated liposomal doxorubicin inhibits topoisomerase II, selectively damaging cancer cell DNA. The combination has been studied primarily in women with platinum-sensitive recurrent epithelial ovarian cancer, displaying an overall safety and activity profile similar to that of carboplatin plus pegylated liposomal doxorubicin alone. Maintenance with farletuzumab as a single agent has also been explored following combination cycles[1][6][4].
