Overview

This is a **combination therapy** comprising a targeted **antibody–drug conjugate (ADC)** containing **farletuzumab** (a humanized monoclonal antibody directed against folate receptor alpha), **eribulin** (a microtubule dynamics inhibitor, used as the cytotoxic payload), and **oral corticosteroids** (a class of steroid hormones typically administered to manage or prevent infusion or allergic reactions). - **Farletuzumab** targets folate receptor alpha (FRα), which is highly expressed in various epithelial cancers, such as ovarian and endometrial cancers. Its mechanism includes antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity. - **Eribulin** is a synthetic analog of halichondrin B, disrupting microtubule growth and leading to cancer cell apoptosis. - As an ADC (notably MORAb-202/FZEC), farletuzumab is conjugated to eribulin via a cleavable linker, allowing targeted cytotoxic delivery to FRα-positive cancer cells. The inclusion of **oral corticosteroids** is standard practice to reduce the risk of infusion reactions and is not part of the ADC molecule itself. - This combination is being investigated in advanced solid tumors, especially ovarian cancer, triple-negative breast cancer, and some rare gynecologic cancers. - Developed primarily by Eisai.
