Overview

A combination therapy consisting of fasudil (a Rho-kinase inhibitor) and bevacizumab (an anti-VEGF monoclonal antibody) administered via intravitreal injection. This combination targets multiple pathways involved in diabetic macular edema (DME). Fasudil inhibits the Rho/ROCK pathway, which is involved in leukocyte adhesion to vascular endothelial cells and subsequent endothelial damage leading to vessel hyperpermeability. Bevacizumab targets vascular endothelial growth factor (VEGF), reducing abnormal vessel formation and vascular leakage. Clinical studies have shown that this combination provides enhanced and prolonged therapeutic effects compared to bevacizumab monotherapy in patients with center-involving DME, including those refractory to anti-VEGF therapy alone.
