Overview

**Fe0@HMON@DNA-Exo** is a research-stage, biologically logic-gated nano-Trojan-horse platform developed for **triple-negative breast cancer**. It consists of elemental iron synthesized within glutathione-responsive degradable hollow mesoporous organosilica nanoparticles and cloaked with DNA-fragment-containing exosomes derived from SN38-treated 4T1 breast cancer cells. In the high-glutathione, mildly acidic tumor microenvironment, iron release promotes Fenton-reaction reactive oxygen species generation and selective ferroptotic tumor-cell death. The DNA-containing exosomal shell also stimulates cGAS-STING signaling in antigen-presenting cells, promoting innate antitumor immunity. The platform has been reported in preclinical murine TNBC models and is not the same asset as Gate2Brain's G2B-002.
