Overview

Feladilimab + dostarlimab is a **combination therapy** consisting of two monoclonal antibodies: feladilimab, an agonistic anti-ICOS (Inducible T-cell Co-Stimulator) antibody, and dostarlimab, an anti-PD-1 (Programmed Cell Death Protein 1) antibody. Dostarlimab blocks PD-1 to prevent PD-L1/PD-L2 ligation, thereby releasing T-cells from immune checkpoint inhibition to restore antitumor immune responses[1][2][4][6]. Feladilimab, in contrast, stimulates the ICOS pathway to enhance T-cell activation and proliferation. This combination is designed to achieve **synergistic immunotherapeutic effects** in cancer therapy, most notably being investigated in advanced solid tumors and head and neck cancers. The combination is currently in clinical trials, aiming to overcome tumor immune evasion mechanisms by both removing suppression and stimulating proliferation/activation of cytotoxic T-cells[1]. Both drugs are developed by GSK.
