Drug intelligence / Profile preview
fenofibrate + pravastatin + ezetimibe is a triple lipid-lowering combination therapy consisting of the peroxisome proliferator-activated receptor alpha (PPAR-alpha) agonist fenofibrate, the HMG-CoA reductase inhibitor pravastatin, and the cholesterol absorption inhibitor ezetimibe. Developed by Laboratoires SMB, this combination is designed to improve the global atherogenic lipid profile in high-risk patients, such as those with type 2 diabetes, mixed hyperlipidemia, and cardiovascular disease. The regimen has been evaluated as a co-administration of a fenofibrate/pravastatin fixed-dose combination (Pravafenix) with ezetimibe, as well as a single triple fixed-dose combination capsule. By combining three distinct mechanisms of action—PPAR-alpha activation to lower triglycerides and raise HDL-C, HMG-CoA reductase inhibition to reduce hepatic cholesterol synthesis, and NPC1L1 inhibition to block intestinal cholesterol absorption—the therapy provides comprehensive lipid-lowering efficacy.
