Drug intelligence / Profile preview
Fenretinide is a synthetic retinoid derivative (N-(4-hydroxyphenyl)retinamide) developed by CerRx, Inc. for the treatment of various cancers, most notably peripheral T-cell lymphoma (PTCL). Its mechanism of action is distinct from other retinoids; while it can bind to retinoic acid receptors, its primary cytotoxic effect is driven by the induction of reactive oxygen species (ROS) and the accumulation of intracellular ceramide, which leads to apoptosis in malignant cells. CerRx developed a proprietary intravenous emulsion formulation to bypass the absorption limitations and low bioavailability of oral fenretinide. This IV formulation was evaluated in Phase II clinical trials for patients with relapsed or refractory PTCL.
