Drug intelligence / Profile preview
Fenretinide Lym-X-Sorb is an oral lipid-based formulation of the synthetic retinoid fenretinide (4-HPR), specifically designed to overcome the poor aqueous solubility and low bioavailability of the parent compound. Developed primarily for the treatment of pediatric cancers such as neuroblastoma, fenretinide LXS utilizes the Lym-X-Sorb (LXS) delivery system, a patented lipid matrix composed of lysophosphatidylcholine, monoglycerides, and fatty acids that facilitates drug absorption via the lymphatic system. Unlike traditional retinoids that act through nuclear retinoid receptors, fenretinide induces cancer cell apoptosis through receptor-independent mechanisms, including the accumulation of intracellular ceramides and the generation of reactive oxygen species (ROS). Clinical investigations, particularly those led by the South Plains Oncology Consortium (SPOC), have evaluated this formulation in recurrent or resistant neuroblastoma, often in combination with metabolic modulators like ketoconazole to further enhance systemic exposure.
