Ferumoxytol + hydrogen peroxide is a combination of an FDA-approved intravenous iron oxide nanoparticle (ferumoxytol) and the oxidizing agent hydrogen peroxide. While ferumoxytol alone is approved for treating iron deficiency anemia, recent research has demonstrated that when combined with low concentrations of hydrogen peroxide, it acts as a catalytic "nanozyme" therapy. This combination generates reactive oxygen species (ROS) via peroxidase-like activity, leading to targeted disruption and killing of pathogenic bacterial biofilms—especially those formed by Streptococcus mutans—while sparing commensal oral bacteria. The mechanism involves preferential binding of the nanoparticles to biofilm matrices and pathogen-specific glucan-binding proteins, followed by localized ROS generation from catalyzed decomposition of H2O2. This results in effective prevention and treatment of dental caries in preclinical models and clinical studies, as well as eradication of chronic endodontic biofilm infections without significant adverse effects or cytotoxicity. Additionally, this approach may stimulate regenerative pathways in dental stem cells[1][2][3][4][5][6][7][9].
