Overview

This is a combination cell therapy consisting of fetal mesenchymal stem cells (MSCs) and fourth-generation safety chimeric antigen receptor (4SCAR) T cells. Fetal MSCs are multipotent stromal cells derived from fetal tissue, known for their immunomodulatory and regenerative properties. 4SCAR T cells are genetically engineered T lymphocytes expressing a fourth-generation CAR construct, which includes an inducible caspase-9 suicide gene for enhanced safety. The CAR targets B-cell and plasma cell surface antigens such as CD19 and BCMA, enabling the modified T cells to recognize and kill pathogenic B or plasma cells. The combination aims to leverage the immunoregulatory effects of MSCs with the targeted cytotoxicity of CAR-T therapy, potentially improving efficacy while reducing toxicity in autoimmune diseases[7]. Early clinical studies suggest this approach may be safe and feasible for treating connective tissue diseases.
