FG-3019 + entecavir is an investigational combination therapy studied for the treatment of chronic hepatitis B virus (HBV) infection with moderate to severe liver fibrosis[1]. FG-3019 (also known as pamrevlumab) is a fully human monoclonal antibody that targets and inhibits connective tissue growth factor (CTGF), a key driver of fibrotic disease[1][2][5]. Entecavir is a nucleoside analog and potent inhibitor of HBV DNA polymerase, used as first-line antiviral therapy for HBV, owing to its low resistance profile[1]. The combination aims to suppress HBV replication (via entecavir) and actively reverse liver fibrosis (via FG-3019)—addressing both the viral cause and its fibrotic consequences. FG-3019 acts by binding CTGF, thereby interfering with fibroblast proliferation, myofibroblast differentiation, and extracellular matrix deposition, crucial steps in liver fibrosis[1][4]. FibroGen, which developed FG-3019, has advanced its clinical development targeting connective tissue diseases, and is running phase 2 trials of this combination for HBV-related liver fibrosis[1][5].
