Overview

A combination therapy consisting of **figitumumab**, a fully human IgG2 monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R); **cisplatin**, a platinum-based chemotherapy agent; and **gemcitabine**, a nucleoside analog chemotherapy. Figitumumab blocks IGF-1R autophosphorylation and inhibits downstream signaling, suppressing tumor proliferation and survival. The combination aims to enhance the cytotoxic effects of chemotherapy by disrupting both growth/survival pathways (IGF-1R) and DNA synthesis (cisplatin/gemcitabine). This regimen has been studied in various cancers, including gastrointestinal cancers and non-small cell lung cancer, although large-scale phase III studies, particularly in NSCLC, have failed to demonstrate additional efficacy with the combination and have reported increased toxicity[1][2][3][4][6][7].
