**Fimaporfin + poly-ICLC** is an investigational combination for vaccine enhancement and immunotherapy, utilizing fimaporfin (a photosensitizer, also known as TPCS2a or Amphinex) and poly-ICLC (a Toll-like receptor 3 agonist, marketed as Hiltonol). Fimaporfin is used in photochemical internalization (PCI) technology to improve the cytosolic delivery of antigens within antigen-presenting cells via localized light activation, thereby strongly enhancing MHC class I presentation and potentiating CD8+ T-cell responses during vaccination. Poly-ICLC is an immune adjuvant that mimics viral double-stranded RNA to activate innate immunity through TLR3, further enhancing adaptive immune responses. The combination has shown to be safe, with dose-limiting local effects primarily for fimaporfin doses above 17.5 µg, and can significantly boost antigen-specific CD8+ T-cell and humoral responses when used as part of vaccine regimens, notably for cancer and infectious disease vaccines. Primary indications investigated are cancer immunotherapy (notably solid tumors and HPV-associated malignancies) and vaccine enhancement[1][2][5][7].
