Overview

AL-60371/AL-817 is a fixed-dose combination otic suspension developed by Alcon Research for the treatment of acute otitis media with tympanostomy tubes (AOMT). It consists of finafloxacin (AL-60371), a novel fluoroquinolone antibiotic, and dexamethasone (AL-817), a potent corticosteroid. Finafloxacin is characterized by its enhanced antibacterial activity under acidic conditions, which are typical of infected ear environments. It works by inhibiting bacterial DNA gyrase and topoisomerase IV, enzymes essential for bacterial DNA replication and transcription. Dexamethasone serves as an anti-inflammatory component, reducing the swelling, redness, and pain associated with the infection. The combination was investigated in Phase 2 clinical trials as a single-application treatment, aiming to provide a more convenient alternative to multi-day dosing regimens of existing ototopical treatments like Ciprodex.
