Overview

It also modulates various molecular pathways, inhibits histone deacetylases, and impacts T-cell phenotype for immunomodulation[3]. - **Tacrolimus** is a calcineurin inhibitor that binds to FK506-binding protein 12, forming a complex that inhibits calcineurin, thus blocking T-lymphocyte activation and interleukin-2 transcription. It is primarily used to prevent organ rejection and treat autoimmune disorders[5]. - **Corticosteroids** are hormones (either synthetic or natural) that act as powerful anti-inflammatory and immunosuppressive agents by binding to the glucocorticoid receptor and regulating genes involved in immune and inflammatory responses[2][6]. Their primary indications are for treating acute and chronic inflammatory conditions, autoimmune disorders, and as adjunct immunosuppression in transplantation[2][4][6]. Combination therapy is studied for synergistically suppressing immune responses in severe autoimmune diseases and after organ transplantation, and has been tested in conditions such as refractory IgA nephropathy and spinal cord injury[1][3].
