Flagellin + F1 + V is a recombinant fusion protein vaccine candidate designed to protect against plague caused by Yersinia pestis. The vaccine combines three components: - Flagellin, a bacterial protein from Salmonella typhimurium that acts as an immunostimulatory adjuvant by activating Toll-like receptor 5 (TLR5) and NOD-like receptor C4 (NLRC4), leading to pro-inflammatory cytokine production and enhanced immune responses. - F1 antigen, the capsular antigen of Yersinia pestis. - V antigen, another key virulence factor of Yersinia pestis. The fusion of these antigens aims to induce robust humoral and cellular immunity against plague. Preclinical studies in mice and nonhuman primates have shown that the Flagellin-F1-V fusion protein is effective as a plague vaccine. Phase I clinical trials demonstrated dose-dependent immunogenicity and good tolerability at all tested doses[2][4]. The product was initially developed by the National Institute of Allergy & Infectious Diseases (NIAID)[1].
