Overview

NXL103 is an oral, semisynthetic antibiotic combination consisting of two streptogramins—flopristin (streptogramin A) and linopristin (streptogramin B)—formulated in a 70/30 ratio. Developed by Novexel, NXL103 acts as a protein synthesis inhibitor by binding to the bacterial ribosome, with its two components working synergistically to inhibit bacterial growth. It demonstrates potent bactericidal activity against a broad spectrum of Gram-positive bacteria—including methicillin-resistant *Staphylococcus aureus* (MRSA), vancomycin-resistant enterococci (VRE), and *Streptococcus pneumoniae*—as well as some Gram-negative and anaerobic organisms. NXL103 was primarily developed for the treatment of community-acquired pneumonia and complicated skin and soft tissue infections such as acute bacterial skin and skin structure infections (ABSSSI). Clinical trials showed efficacy comparable to amoxicillin for community-acquired pneumonia, with gastrointestinal intolerance being the most common adverse event[1][3][4][5][6][7].
