FLOT + sintilimab

FLOT + sintilimab is a combination therapy that pairs the FLOT chemotherapy regimen (fluorouracil, leucovorin, oxaliplatin, and docetaxel) with sintilimab, a PD-1 inhibitor. This combination is primarily used as neoadjuvant treatment for HER2-negative locally advanced gastric or gastroesophageal junction (G/GEJ) adenocarcinoma. The FLOT component works by targeting rapidly dividing cancer cells through multiple mechanisms, while sintilimab enhances anti-tumor immune responses by blocking the PD-1 pathway. Clinical studies have shown encouraging efficacy with manageable safety profiles, with pathological complete response rates of approximately 17.2% and major pathological response rates of 55.2% in phase II trials.
