Overview

fluconazole + vismodegib is a combination of two small-molecule pharmaceutical drugs. Fluconazole is an antifungal agent that acts mainly as a moderate inhibitor of cytochrome P450 enzymes (notably CYP2C9 and CYP3A4), while vismodegib is an antineoplastic agent that inhibits the Hedgehog signaling pathway by targeting the Smoothened receptor. Co-administration leads to moderate increases in vismodegib exposure (~31% higher AUC at steady state), attributed to inhibited metabolism of vismodegib by fluconazole. No clinically significant increase in adverse events or toxicity has been observed when these drugs are used together in healthy subjects[1][2]. The combination itself is not a distinct commercial product but represents a clinically-studied co-administration scenario.
