Overview

This therapeutic regimen consists of autologous anti-CD19 chimeric antigen receptor (CAR) T-cell therapy preceded by fludarabine-based lymphodepletion chemotherapy. Developed by Union Hospital, Tongji Medical College, Huazhong University of Science and Technology, the treatment targets CD19-positive B-cell malignancies. The CAR-T cells are engineered to recognize and eliminate B cells expressing the CD19 antigen. Fludarabine is administered as part of a lymphodepleting conditioning regimen (often in combination with cyclophosphamide) to create a favorable environment for CAR-T cell expansion and persistence by reducing endogenous T cells and immunosuppressive cells. This specific protocol is being evaluated in clinical trials for relapsed or refractory B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL), Burkitt lymphoma, and follicular lymphoma, sometimes in combination with BTK inhibitors like ibrutinib or zanubrutinib.
