Overview

Fludarabine + cyclophosphamide is a combination chemotherapy regimen primarily used in the treatment of B-cell chronic lymphocytic leukemia (CLL). Fludarabine is a purine analog antimetabolite that inhibits DNA synthesis by interfering with DNA polymerase alpha, ribonucleotide reductase, and DNA primase. Cyclophosphamide is an alkylating agent that crosslinks DNA strands and prevents cell division. The combination increases complete remission rates and reduces minimal residual disease compared to fludarabine alone. This regimen is often used as part of multi-drug protocols for CLL and other hematologic malignancies[2][5][8].
